In-Fusion PCR Cloning Primer Design Function
Currently, only the primer design function is installed. PCR cloning using this primer cannot be performed on the IMC.
What is In-Fusion primer design?
In-Fusion PCR cloning is a method that does not require end blunting or ligation, and allows PCR fragments to be cloned into any vector in one reaction. Efficient cloning is possible by adding the terminal sequence of a linearized vector to the primer of the DNA fragment to be inserted.
How to use In-Fusion primer design
- Load any vector sequence into the feature map.
- Click the "In-Fusion" button.
- The "In-Fusion Primer Design Dialog" will be displayed.
- Specify the forward primers you have already created in "Primer 1" and "Primer 2".
- Specify the restriction enzyme recognition site in the MCS of the vector in "Restriction Site".
- Click "Run".
- The designed "In-Fusion" primer set will be displayed.
- Specify the generated primer set and click "Save as a (New) Primer..." to register this set in the "Primer List".
- Select the generated primer set and click "CSV" to save it as a file that can be read in Excel.
