IMC i01C Restriction Enzyme Digestion of DNA Sequence and Load Fragment

Digestion of the current DNA sequence with restriction enzyme and loading the restriction fragments.

Operation

  1. Load the DNA sequence file.
  2. For example, load "Bsub_50 kb.gbk".
  3. The feature map is displayed.
  4. Select "Cloning -> Restriction Enzyme -> RE Registration & Editing" from the menu.
  5. The "Enzyme Selection" dialog is displayed.
  6. Click the "Select All" button.
  7. All restriction enzymes are checked.
  8. Click the "Show Recognition Site" button.
  9. The Recognition Site window is displayed.
  10. The restriction enzyme name and the number of recognition sites detected are listed in the left pane.
  11. Check with restriction enzyme (multiple selection is possible).
  12. The recognition site of the selected restriction enzyme is listed in the right pane.
  13. Click "Select All".
  14. All check boxes in the right pane are checked.
  15. Click "Digestion ...".
  16. A "Digestion?" Confirmation message will be displayed.
  17. Click "Yes (Y)". 
  18. The "Digestion List" dialog will be displayed.
  19. In this list, file name automatically appended to digestion cut piece, base length, position on original DNA sequence, name of restriction enzyme site at both ends, actual recognition site sequence, DNA sequence and the like are displayed I will.
  20. Click the "Select All" button.
  21. All fragments are checked.
  22. Click "Load ...".
  23. A completion message is displayed.
  24. Restriction fragments are loaded into the current main directory.
  25. Click on one of the loaded nodes.
  26. A feature map of restriction enzyme digested fragments is displayed on the main feature map.
